چکیده

According to MARKEES AND MEYER 1 an increased content of pyruvate is found in the blood of patients in diabetic coma. I t decreased by intravenously injecting thiamine pyrophosphate, but not, however, by injecting thiamine. The authors concluded that in this condition phosphorylation is inhibited. We have found, however, that in these investigations the pyruvate determinations, which were performed according to the method of FRIEDEMANN AND HAUGEN z, have been carried out in such a way that pyrnvate was determined together with acetyl acetate. Overnight storage of the deproteinized blood in the refrigerator, as prescribed by FRIEDEMANN AND HAUGEN and as had been done by MARKEES AND MEYER, did not appear to be efficacious for removing the acetyl

Thiamine Ameliorates Diabetes-Induced Inhibition of Pyruvate Dehydrogenase (PDH) in Rat Heart Mitochondria: Investigating the Discrepancy Between PDH Activity and PDH E1 α Phosphorylation in Cardiac Fibroblasts Exposed to High Glucose

The activity of pyruvate dehydrogenase (PDH) is reduced in diabetic patients. Phosphorylation of the PDH E1 α subunit by PDH kinase contributes to the suppression of PDH activity. PDH requires thiamine as a coenzyme. We investigated the exact mechanism of diabetes-induced PDH inhibition, and the effect of thiamine in both in vivo and in vitro experiments. Treatment of rats with thiamine signifi...

Thiamine and Cocarboxylase Concentration

In patients with congestive heart failure, thiamine deficiency has been demonstrated by the test-dose method (1, 2), by measurement of the lactate-pyruvate ratio of the blood (3), and by determination of blood cocarboxylase (4). The oxidation of pyruvate to acetate requires thiamine in the form of cocarboxylase (thiamine pyrophosphate). Olson, Pearson, Miller, and Stare have shown that thiamine...

The Effect of Endurance Exercise on mTORC1 Marker Pathway in the Soleus Muscles of Type 2 Diabetic Rats

Background mTORC1 marker pathway is one of the crucial pathways for the regulation of transcription level and an essential route involved in protein synthesis in skeletal muscles. Objective This study aimed to investigate the effect of endurance training on mTORC1 marker pathway in soleus muscle of type 2 diabetic rats. Methods In this experimental study, 16 Sprague-Dawley male rats (Mean±SD ...

Structural basis for inactivation of the human pyruvate dehydrogenase complex by phosphorylation: role of disordered phosphorylation loops.

We report the crystal structures of the phosporylated pyruvate dehydrogenase (E1p) component of the human pyruvate dehydrogenase complex (PDC). The complete phosphorylation at Ser264-alpha (site 1) of a variant E1p protein was achieved using robust pyruvate dehydrogenase kinase 4 free of the PDC core. We show that unlike its unmodified counterpart, the presence of a phosphoryl group at Ser264-a...

Antihyperglycemic, Antihyperlipidemic and Antioxidant Effect of Phyllanthus rheedii on Streptozotocin Induced Diabetic Rats

The aim of this study was to investigate the effects of ethanolic extract of the whole plant of Phyllanthus rheedii wight P.rheedii as antihyperglycemic, antihyperlipidemic and antioxidant effect in streptozotocin (STZ) induced diabetic rats. Male Wistar rats were administered P.rheedii (250 mg/kg) orally for 21 days and blood glucose level was measured weekly. At the end of 21 days, the serum ...
